A selective technique for the isolation of Neurospora crassa from soil.

چکیده
Neurospora is among the most studied of all eukaryotes. It is a convenient organism for the study of genetic polymorphism through analysis of isozymes (Yu, Garrett & Sussman, 1971 ; Reddy & Threlkeld, 1971,1972), and the possibility of population studies with Neurospora has been demonstrated by Perkins (1970) who collected isolates from over 60 localities in the tropics. However, Perkins' collection was derived from visibly growing colonies on vegetation or upon the soil surface in burned areas, so selection of certain forms may have occurred. To be more representative of the potential genetic variability in nature, collections should include samples from a diversity of soil habitats. We report here on a technique found effective for the isolation of Neurospora crassa from soils in India.

Biotransformation of Hydrocortisone by Neurospora crassa
The ability of Neurospora crassa FGSC 4335 in the biotransformation of hydrocortisone was investigated. The microorganism produced two major metabolites after incubation with the substrate for seven days. Each microbial product was purified chromatographically and identified on the basis of spectral data. The products were identified as 11?,17?,20?,21-tetrahydroxypregn-4-en-3-one (II) and 11?-h...
متن کاملThe dicyclohexylcarbodiimide-binding protein of the mitochondrial ATPase complex from Neurospora crassa and Saccharomyces cerevisiae. Identification and isolation.
Incubation of mitochondria from Neurospora crassa and Saccharomyces cerevisiae with the radioactive ATPase inhibitor [14C]dicyclohexylcarbodiimide results in the irreversible and rather specific labelling of a low-molecular-weight polypeptide. This dicyclohexylcarbodiimide-binding protein is identical with the smallest subunit (Mr 8000) of the mitochondrial ATPase complex, and it occurs as olig...
متن کاملIsolation of Neurospora crassa bradytrophs.
A method was developed for the isolation of Neurospora bradytrophs. The bradytrophs (representing lesions in a number of pathways) were resistant to DL-p-fluorophenylalanine when growing in a leaky fashion but were sensitive when grown in the presence of their stimulating supplement.
متن کاملIntracellular localization, isolation and characterization of two distinct varieties of superoxide dismutase from Neurospora crassa.
1. Neurospora crassa was found to contain two distinct superoxide dismutases. 2. Most of the activity is associated with the cytosolic fraction and was shown to be the Cu/Zn-containing form of the protein. 3. Mitochondria isolated from Neurospora crassa showed two distinct superoxide dismutases: a cyanide-sensitive Cu/Zn-containing protein and a cyanide-insensitive form which probably contains ...
متن کاملIsolation and characterization of low-kynureninase mutants of Neurospora crassa.
Two kynureninase activities are known in Neurospora crassa, one of which (kynureninase I) is inducible, the other (kynureninase II) being constitutive. A method is described for the isolation of low-kynureninase mutants of N. crassa. When grown on an inducer, the mutants show significantly less kynureninase I activity compared with wild type, whereas constitutive kynureninase II activity is una...
